The overall aim of this observational cohort study is to monitor the safety of Mpox
vaccines in a cohort of subjects in DRC to detect safety signals
Primary objectives
Secondary objectives
Study period: 3 months after the last dose of one of the Mpox vaccines.
Follow-up: All participants completed questionnaires through telephone calls at weekly intervals in the first month (D7, D14, D21 and D28, with D0 being the day of vaccination), and once monthly for the two remaining months (D56 and D84) after each vaccine dose administration. Participants in the reactogenicity subset were followed up at additional timepoints in the first 14 days after each dose (D1, D4, D7 and D14, with D0 being the day of vaccination).
Data collection: On a smartphone using a mobile app (ODK Collect). Sample size: 30,000 participants (with a reactogenicity subset of 1,000 per vaccine).

Project duration
May 2025 to December 2025 (Duration of participant enrollment)
The last enrolled participant was followed up until Day 81 after vaccination.
Research area
Target population
Participants were recruited among Mpox vaccinees, vaccinated at the vaccination sites participating in this CEM study. The planned sample size was 30,000.
The actual sample size has 24,218 (after database cleaning) following the decision made in December 2025 to halt the enrolment of new participants, given the low likelihood of Mpox vaccine deployments in the near future and the significant decline in Mpox case numbers.
Geographical scope
13 health zones/districts across 3 provinces (Kinshasa, Equateur and Sankuru) of the Democratic Republic of Congo.
Study design
An observational prospective single-arm longitudinal study in selected vaccination centres in the Democratic Republic of Congo.
